富含血小板血浆对组织工程脂肪移植效果的影响(1)

     [摘要]目的:在移植的组织工程脂肪组织中加入含激活剂的富含血小板血浆(PRP)或不含激活剂的富含血小板血浆,通过观察脂肪组织各个时间段的成活率来证明PRP的作用。方法:根据Landesberg二次制备法制备PRP。按1000U凝血酶溶于1ml质量百分比为 10%的CaCl2配制激活剂。从兔背部取脂肪组织制备好,按实验分组分别加入不同的成分移植到兔子耳部。分别在术后1月、3月、6月测量体积变化。结果:含激活剂的PRP加入组织工程脂肪移植后3个时间段的体积成活率均高于对照组。结论:加入激活剂的PRP释放生长因子增多,释放速度增加,间接促进了脂肪干细胞分裂、分化、成活,促进了移植脂肪组织的更新,使脂肪移植成活率增加。

    The effects of platelet-rich plasma (PRP) upon the fat tissue after grafts

    Abstract: Objective To explore the PRP's role in adipose tissue transplantation by detecting the survival rate of adipose tissue at each time point. Methods PRP was prepared according to the method of Landesberg.The activator was made up of l000U bovine thrombin and 1ml 10% calcium chloride solution. The fat tissues taken from the rabbit back were grafted to the rabbit ears in different experimental groups. The volume changes were measured after 1 month,3 months,6 months,respectively. Results After transplantation,the survival rates of the group within the activator at each time were higher than the group without the activator. Conclusion The activator promoted the the survival rate of transplanted adipose tissue. The reason was that the PRP with the activator released more growth factors,which contributed to adipose stem cells' division,differentiation,and survival.

    近年来,软组织缺损的治疗随着科学技术的发展而出现了许多新的技术,脂肪移植就是其中比较重要的技术[1],但单纯的脂肪组织移植却有着很不稳定的吸收率,甚至可以高达90%。组织工程的出现,为脂肪移植提供了新的思路,我们课题组针对此进行了自体脂肪颗粒(adipose granu1e,AG)复合血管基质成分(stromal vascular fraction cells,SVF)移植的实验[2],虽然取得了一定的效果,明显提高了脂肪组织的成活率 ......
